We are seeking a highly motivated and experienced scientist specializing in recombinant protein production to join our dynamic research team and accelerate our Cryo-Electron Microscopy and Crystallography workflows. The successful candidate will play a pivotal role in the expression, purification, and characterization of recombinant proteins and complexes for structural studies supporting small molecule and biologics drug discovery. This position requires a deep understanding of protein chemistry for structural and biophysical studies, great wet lab skills, and a team-oriented approach to delivering high-quality samples within project timelines. In addition, the candidate would require experience managing a protein production laboratory and external collaborations/CRO's, and the ability to oversee both scientific and operational aspects of the lab to support a diverse portfolio of projects.
The ideal candidate will have a strong background in protein expression in bacterial, insect and mammalian systems and excellent protein purification skills using standard techniques for cryo-EM and crystallography, as well as for reagent generation for assays.
Key Responsibilities:
• Protein Expression: Transfection of plasmids into mammalian cells to express key viral proteins.
• Protein Purification: Design, optimize, and execute protocols for the purification of recombinant proteins using various chromatography techniques (e.g., affinity, ion exchange, size exclusion).
• Biophysical Characterization: Perform biophysical characterization of purified proteins, including but not limited to, SDS-PAGE, analytical SEC, and Differential Scanning Fluorimetry (DSF).
• Data Analysis & Reporting: Maintain detailed records of experimental designs, data, and results. Analyze and interpret data for presentation to key stakeholders.
• Team Collaboration: Collaborate closely with design team to determine protein constructs with optimal stability and functionality.
Qualifications (Minimal acceptable level of education, work experience, and competence)
• Advanced degree in chemistry, structural biology, biochemistry or related field with relevant experience in biotech or large Pharmaceutical company.
• Proven experience supporting drug discovery programs using structural biology and biochemistry.
• Good communication and interpersonal skills.
• Proven experience working with matrix teams
• Self-driven, creative and entrepreneurial with ability to support multiple programs simultaneously.
• Cryo-EM and Crystallography experience highly is desirable, but not required.
• Develop and implement short and long-term strategies to enhance protein production capabilities.
• Provide guidance on cloning, protein expression, purification, and biochemical and biophysical characterization
• Help troubleshooting recombinant protein expression and purification
• Protein engineering experience is desirable but not required
• May train, mentor, and supervise a team of scientists, technicians, and support staff
• Membrane protein experience is desirable but not required
• Knowledge of protein prediction and computational approaches for protein design
• Stay current on emerging technologies and instrumentation in protein chemistry and biochemistry to introduce innovative approaches to the facility
• Troubleshoot and optimize workflows to improve the efficiency and reproducibility of experimental results.
