GENERAL SUMMARY/ OVERVIEW STATEMENT: The Research Technician will perform standard laboratory procedures in the lab of Dr. Gian Paolo Dotto at CBRC/MGH. This include routine molecular biology, histology/immunochemistry and/or flow cytometry in support of projects in cutaneous biology and skin cancer. Other duties might involve mouse genotyping, euthanasia and tissue harvest. This position also requires record keeping via an ELN, laboratory plasmids/cell database management, ordering and maintenance of lab reagents stocks. PRINCIPAL DUTIES AND RESPONSIBILITIES: * Supports ongoing projects. This may require perform tissue culture, primary skin fibroblasts and primary keratinocyte preparation, preparation of lentiviral stocks for gene manipulation (overexpression, shRNA and CrispR). Preparation of plasmid maxipreps. Performing tissue collection and processing, histology and immunohistochemistry, RNA and DNA extraction, cDNA synthesis and PCR.
* Maintain laboratory stocks of reagents, ordering.
* Conduct analysis of results in consultation with PI.
* Attend weekly laboratory meetings and maintains laboratory electronic notebooks.
* Will perform other duties as assigned. SKILLS/ABILITIES/COMPETENCIES REQUIRED: * Must exhibit initiative and be able to work independently when trained.
* Must be accurate and efficient in performance of routine experiments.
* Ability to work in a group environment and be of assistance to a number of research projects.
* Good written and verbal communication skills. EDUCATION: Bachelor of Science or Bachelor of Arts required. EXPERIENCE: Lab experience preferred . Molecular biology and/or tissue culture experience is desired but not required. Will train in more advanced techniques.
