Cogent's lead therapeutic candidate, bezuclastinib (CGT9486), is a precision kinase inhibitor designed to selectively and potently inhibit the KIT D816V mutation. This mutation is responsible for driving a rare and serious condition called Systemic Mastocytosis which can severely impact many different tissues and organs in the body. We are also studying CGT9486 to treat advanced gastrointestinal stromal tumors (GIST), which have a strong dependence on oncogenic KIT signaling.

The role:

Cogent Biosciences is growing! This position would be an integral part of our research team in Boulder, CO. The Senior Scientist will be responsible for designing and executing cell-based assays to support target validation and project advancements in close collaboration with other Discovery Biology team members. The ideal candidate will have a strong understanding of oncogenic and inflammatory pathways and will use his/her knowledge and experience to build cell-based assay systems to evaluate desired molecular interactions to support SAR screening. This individual will have a seat at the table where they could intellectually contribute and work with a multi-disciplinary team towards the common goal of bringing forward innovative cancer therapies to patients with unmet needs.

What You Will Do:
  • Extensive mammalian cell culture expertise to develop, optimize, execute, and troubleshoot robust cell-based assays to support ‘Hit to Lead' discoveries.
  • Design, troubleshoot, and execute various in vitro cell based assays to validate novel targets (eg. HTRF, AlphaLISA, Mesoscale and Chemiluminescence).
  • Engineer stable cell lines for in vitro functional assays and in vivo studies (CRISPR experience is a plus).
  • Operate, implement, and troubleshoot laboratory instruments and automations.
  • Perform comprehensive data analysis, curve fitting to generate and report weekly screening results to drive SAR.
  • Work cross-functionally with enzymologists, structural biologists, medicinal chemists and pharmacologists to inform program decisions.
  • Execute experiments with rigor and clearly present findings at project team meetings.
  • Champion emerging technologies and methods to serve program needs more efficiently.
  • Actively stay up to date on scientific advancements from the literature.
  • Work "hands on" in the lab to execute the assays above as well as other collaborative projects with team members.


What You Bring:
  • MS, BS, or PhD in Cell Biology, Molecular Biology, or Biochemistry/Biophysics, with 5-10 years (for BS/MS) and 2+ years (for PhD) of biopharmaceutical experience. (Level will be commensurate with experience).
  • Strong interpersonal and verbal/written communication skills.
  • Experience working on cross-functional drug discovery teams.
  • Expertise in cell culture techniques across a variety of cell lines (cancer cell line experience preferred) and assay endpoints (phospho inhibition, proliferation, 3D, 2D assays, etc.
  • Substantial experience using standard molecular and cellular biology assay methods (i.e., ELISA, Mesoscale, HTRF, Western blotting, In-cell Western, Flow Cytometry, BRET, chemiluminescence, DNA preparations, qPCR, RT-qPCR, primer design, cell line generation, etc.).
  • Basic knowledge on data analysis tools (GraphPad Prism, Excel, familiarity with Spotfire, Gene Data, etc.).
  • Good and thoughtful listener with a passion to work at the bench independently with a can-do attitude.
  • Desire to work within a fast-paced, innovative, and collaborative environment with a continual desire to learn.
  • Ability to prioritize responsibilities and work efficiently to meet deadlines.
