Description

This position is responsible for cloning and protein expression construct design to support antibody discovery screening programs. Additional responsibilities include sequence analysis, library construction, and phage display screening procedures.

Essential Functions
  • Design and perform cloning methods, PCR methodologies, and vector and primer design
  • Vector optimization for libraries, and creation and construction of the libraries themselves
  • Design, optimization and implementation of RT-PCR from single cells and tissues
  • Analyze DNA sequences
  • Analyze sequence activity relationships to enable affinity maturation of antibodies
  • Perform DNA mini-, midi-, maxi-prepping procedures
  • Maintain and archive DNA clones, and bacterial glycerol stocks
  • Prepare and process samples for next generation sequencing (NGS) and run them on our Illumina instrumentation
  • Optimize cloning and nucleic acid analysis workflows, including library preparation and analysis
  • Assist with phage display library generation, panning, reformatting, and sequencing as needed.
  • Organize, summarize and present data and results at department and project team meetings
  • Assist/mentor junior, and new, laboratory personnel
  • Develop an expanding technical and theoretical knowledge base in molecular biology with a growing understanding of the scientific literature in relevant areas of research
  • Maintain a detailed and well-organized record of experiments in an electronic notebook
  • Maintain familiarity with literature to propose and implement improved procedures and protocols, and to aid in problem solving
  • May prepare technical reports, summaries and detailed protocols, and contribute to journal articles and company reports as requested
  • Follow and adhere to SOPs and laboratory safety regulations

Requirements

Qualifications

Education & Experience:

· Theoretical knowledge typically achieved through a related four-year college experience, such as a Bachelor's degree in Molecular Biology or related scientific field

· Five years of molecular biology techniques, such as molecular cloning, PCR, E. coli transformation and cultivation, DNA extraction, enzyme digestion and gel electrophoresis

Knowledge and Competencies:

· Experience in cloning and PCR methodologies including restriction digests, Gibson assembly, vector and primer design

· Working knowledge of protein expression vectors (promoter, transcriptional signals, etc.) and have the ability to analyze DNA and protein sequences

· Ability to communicate clearly, and to report and present experimental results and analysis to company colleagues

· Effective interpersonal skills, including excellent written and oral communication skills

· Ability to multi-task, organize multiple assignments, and work on several projects simultaneously while adhering to deadlines

· Self-motivated, independent, and results-oriented

· Team player, intensely committed to success and getting the job done well in a highly dynamic environment

· Ability to work independently while also willing to follow protocols

· Strong analytical and organizational skills, with high attention to details
