This position is responsible for therapeutic antibody screening from single B cell sorting and phage display libraries, cloning and expressing antigens, antibodies and their variants for our antibody discovery screening programs. Additional responsibilities include sequence analysis as it relates to antibody affinity maturation and target validation.
Essential Functions
· Design and implementation of optimized antibody screening strategies from human and mouse single B cells
· Construction of natural and synthetic antibody libraries for mammalian cell and phage display
· Design and implementation of phage display and mammalian cell display selection strategies
· Design, creation and implementation of cloning strategies using different cloning methods, PCR techniques and primer design
· Design, creation and construction of vectors for AnaptysBio libraries
· Design, optimization and implementation of RT-PCR from single cells and tissues
· Analyze and mine DNA sequences from Sanger and NGS
· Analyze sequence activity relationships to enable affinity maturation of antibodies
· Analyze RNA seq data for target validation
· Perform DNA mini-, midi-, maxi-prepping procedures
· Maintain and archive DNA clones, and bacterial glycerol stocks
· Organize, summarize and present data and results at department and project team meetings
· Assist/mentor junior, and new laboratory personnel
· Develop an expanding technical and theoretical knowledge base in molecular biology with a growing understanding of the scientific literature in relevant areas of research
· Maintain a detailed and well organized record of experiments in an electronic notebook
· Maintain familiarity with literature to propose and implement improved procedures and protocols, and to aid in problem solving
· May prepare technical reports, summaries and detailed protocols, and contribute to journal articles and company reports as requested
· Follow and adhere to SOPs and laboratory safety regulations

Knowledge Requirements:
· Practical knowledge of phage display
· Extensive experience with protein expression and display vectors (promoter, transcriptional signals, etc.) and have the ability to analyze DNA and protein sequences
· Proficiency working with DNA analysis software programs such as Sequencher, DNAStar or their equivalent, and with Microsoft Office (Word, Excel, Powerpoint)
· Ability to recognize when it is appropriate and necessary to modify a standard procedure or protocol
· Consistent success in independently interpreting, and summarizing results, and in proposing next steps
Personal Competencies:
· Ability to communicate clearly, and to report and present experimental results and analysis to company colleagues
· Effective interpersonal skills, including excellent written and oral communication skills
· Ability to multi-task, organize multiple assignments, and work on several projects simultaneously while adhering to deadlines

Requirements
Education & Experience:
- Theoretical knowledge typically achieved through post-graduate college experience, a PhD in Molecular Biology is preferred
- 3+ years molecular biology experience.
