Scientist in Synthetic Biology

Job Description

We are seeking multiple full-time Scientists, who are highly experienced in molecular biology, DNA synthesis or assembly techniques, or emulsion-based chemistry. You will join our rapidly growing scientific team at Replay to design, engineer, and implement the next generation of virus-based gene therapies and synthetic biology platform technologies!

Replay is a start-up, VC-backed biotechnology company located in San Diego, CA focusing on developing gene therapy modalities using herpes simplex virus (HSV) as a vector for gene delivery and leveraging synthetic biology techniques, like DNA synthesis and assembly, to improve vector engineering.

This position will give you the opportunity to take the lead in further developing and implementing virus genome engineering with multiple DNA assembly platforms, to deliver critical support to projects within the company. An ideal applicant has extensive experience in molecular biology or chemistry, a desire to develop synthetic biology technologies, and creative ideas about DNA assembly processes.

We are looking for a Scientist to:

· Implement multiple DNA assembly methods to construct gene therapy candidates.

· Perform experimental planning, design and analysis.

· Develop and carry out Replay's scientific goals.

· Manage a small laboratory team.

· Provide laboratory support and carry out lab-based molecular biology experiments.

· Maintain organized records of group progress and scientific data.

To meet these challenges, you will need to:

· Hold a PhD in a relevant area of Biological or Chemical Sciences and/or 5-10 years of relevant experience.

· Have experience in (some of the following):

o Molecular biology techniques, especially in gene cloning and DNA assembly techniques, e.g. transformation-associated recombination (TAR), Gibson, Golden Gate, BioBrick, PCA, and others.

o Microbiology, including E. coli and/or yeast

o Gene synthesis chemistry and optimization, emulsion chemistry and/or microfluidics

· Be team-oriented, motivated, and organized.

Skills desired for Synthetic Biology Scientist

Successful candidates will possess some of the following specific skills:

Effective communicator and manager, team-player, creative, organized and detail-oriented.

Lab skills: Gene synthesis, emulsion-based chemistry, microfluidics, DNA assembly (Gibson or yeast recombination), DNA synthesis, virus engineering, yeast biology, methods for high-molecular weight DNA, bacterial and yeast growth and transformation, PCR, nucleic acid extraction, electrophoresis, including CHEF/FIGE, vector sequence design, cloning, primer design, tissue culture, virology (especially with herpesviruses), molecular genetics studies, CRISPR or other editing

Computer skills: DNA sequence programs (i.e., ApE, SnapGene, Geneious), Microsoft Office, email, Zoom, electronic lab notebooks. Computational/bioinformatic skills.
