POSITION OBJECTIVES:
  • The Therapeutic Generation and Screening (TGS) team of Takeda's Global Biologics Department is seeking an energetic and creative scientist with demonstrated skills in molecular biology and cell culture.
  • The successful candidate will divide their time between designing DNA constructs and generating stable cell lines to be used in screening assays for binder selections for various therapeutic modalities.


POSITION ACCOUNTABILITIES:
  • Design DNA constructs for protein expression of antibodies, single domains, multispecifics, enzymes, membrane proteins, etc. or for lentiviral transduction in stable cell line generation or chimeric antigen receptor expression.
  • Transfect or transduce cell lines to generate overexpression stable cell lines.
  • Design CRISPR constructs to engineer knock-outs or knock-ins in stable cell lines.
  • Characterize these cell lines using assays such as flow cytometry, Western blot, PCR, etc.
  • Work collaboratively and effectively in a multi-disciplinary team environment as a team player, yet independently manage projects and lab work.
  • Multitask and work independently to meet aggressive timelines.
  • Independently design, develop and execute research assignments and contribute to multiple projects, in parallel.


DIMENSIONS AND ASPECTS:

Technical/Functional (Line) Expertise
  • Familiarity with nucleotide sequences such as promoters, Kozak sequence, and IRESs and regulatory elements and other protein domains. Familiarity with backbone vectors such as pcDNA3.4 and common lentiviral vectors.
  • Proficiency and hands-on experience with mammalian cell culture of both adherent and suspension cell lines, transduction and transfection methodologies, Vi-Cell, Octet, SDS-PAGE, Western blot, flow cytometry and production and selection of stable cell lines.
  • Familiarity with chimeric antigen receptor structure and function. Experience with culturing of CAR-T and NK cells is a plus.
  • Experience with CRISPR design and gene editing for both knock-out and knock-in of mammalian cell lines and molecular biology techniques such as primer design, DNA/RNA isolation, PCR etc.
  • Familiarity with databases at Uniprot and NCBI, sequence analysis using BLAST and alignments, and DNA software such as Snapgene.
  • Experience in dealing with managing budgets and working with CROs for gene synthesis
  • Familiarity with Genedata and DNA inventory systems is a plus.

EDUCATION, BEHAVIORAL COMPETENCIES AND SKILLS:
  • PhD degree in a scientific discipline, or
  • MS with 6+ years experience, or
  • BS with 8+ years experience
