We are seeking an experienced individual to work as a Biologist at the National Institutes of Health in Bethesda, MD.

This is a long-term contract position which offers:

*TASKS. *The contractor shall:

* Culture bacterial and fungal strains for in vivo infections and in vitro uptake assays.

* Handle radioactive material involved in the in vitro uptake assays such a tritium and C14

* Validate imaging data with various post-necropsy assays using techniques such as autoradiography, microscopy, flow cytometry, immunoblotting, qPCR

* Perform binding assays and ELISA to test the affinity of novel ligands to specific microbial targets.

* Perform radioTLC and other equivalent assays to ascertain the metabolic reactions and by-products produced in the microbes

* Participate in the planning and performing of research studies

* Request and maintain the necessary supplies for laboratory imaging use; and

* Develop new methods and procedures or substantially modify existing guidelines to resolve technical problems

* Perform various in vitro and ex vivo functional assays that include: growth and maintenance of microbial cultures

* Isolation and purification of microbial components such as proteins and polysaccharides for the development of PET tracers; in vitro uptake and binding assays with newly developed PET tracer prototypes

* Inoculating rodents (mice or rats) with infectious agents

* Validating the imaging results with a complementary ex vivo experimental techniques such as PCR, immunoblotting, light and immunofluorescence microscopy and ELISA; data collection and analysis

*REQUIREMENTS.* The contractor must have:

* Master's Degree in Microbiology or other related discipline. Three (3) years of specialized experience plus a BS degree is equivalent to a Master's degree.

* Minimum of Three (3) years of experience at Masters level

* Minimum of Five (5) years' experience at BS/BA level

* Technical knowledge of aseptic techniques in culturing microbial strains and experience in the field of mycology

* Knowledge of fungal metabolic pathways and pathogenesis

* Ability to isolate various fungal cell wall components, other proteins/biomolecules of interest, and characterize whole cell lysates for metabolic products

* Technical expertise in various molecular assay techniques like western blotting, ELISA, PCR, fluorescence microscopy and flow cytometry

* Technical knowledge of developing rodent models of fungal infection

* Knowledge of basic animal handling procedures such as anesthesia use, monitoring of animal vital signs, tissue harvesting and euthanasia

* Knowledge in developing new assays, troubleshooting procedures and equipment, validating test results with the use of appropriate controls, record and analyze data
