About Scientist, T Cell Biology - Lexington, MA

Our Alloy Discovery Services group is looking for a Scientist to join our team in Lexington, Massachusetts. The position will report directly to our Director of External Discovery & Innovation and work with our Discovery Services team to support the discovery and functional characterization of antibody-based therapeutics, effector cell engagers, chimeric receptors, and TCR-based therapeutics. You will be using the latest in cell-based functional assays to develop screening methods for molecules characterized by the best functionality and potency with the ultimate goal of helping others to make better medicines.

Principal Responsibilities
  • Development and optimization of cell-based activity assays for primary and engineered human immune cells (T cells, NK cell, Macrophages)
  • Engineering of CAR T and NK cells using viral vectors or other gene editing approaches
  • Expansion and analysis of genetically modified primary T cells and other immune cells
  • Optimization of gene editing in primary immune cells using the CRISPR Cas9 system
  • Utilize an array of analytical and molecular techniques, next generation sequencing, to understand the dynamics and response of genetically modified T cells and other immune cells to antibody-based therapeutics
  • Effectively collaborate within Alloy's team and with external partners to develop study design and obtain test materials and reagents
  • Contribute to study design, execution, and data analysis
  • Compile and present data to functional teams
  • Being generous when others are needy, greedy or fearful

Required Qualifications and Skills
  • PhD or M.S. in cell biology, immunology, or related discipline with 5+ years of experience
  • Experience with mammalian cell culture; isolation, handling, and characterization of primary human immune cells
  • Experience with standard immunological assays (e.g. activation markers, cytokine release, cell killing; flow cytometry, MSD/Luminex and other methods)
  • Experience working with automated HTP liquid handlers is a plus
  • Competency in basic molecular biology techniques, including cell line engineering
  • Working knowledge of various software (e.g. FlowJo, GraphPad Prism, FACSDiva, Geneious)
  • A sense of urgency, speed and persistence
